چکیده

Three diluents were studied, to determine which is the best for the automated immunochemical measurement of specific serum proteins. Nine serum proteins (orosomucoid, a 1-antitrypsin, a2-macroglobulin, haptoglobin, transferrin, C3, lgG, IgA, and 1gM) were measured in physiological saline (9 g NaCI/liter), tris(hydroxymethyl)aminomethane buffer (0.01 mol/liter; pH 7.4), and physiological saline containing polyethylene glycol (‘PEG 6000,” 40 g/liter). Criteria were: reaction rate, analysis rate, carryover between samples, steady-state approximation, precision, and correlation with other methods. Saline containing polyethylene giycol is the best of the three diluents for use in continuous-flow nephelometric analysis of serum proteins.

Optimizing reference values for the measurement of alpha 1-antitrypsin in serum: comparison of three methods.

We studied three methods (rate nephelometry, radial immunodiffusion, and trypsin-inhibitory capacity) for their ability to detect those individuals with a deficiency of alpha1-antitrypsin. The phenotype represented in 170 serum samples was determined by isoelectric focusing as the reference method. All three methods correctly identified Pi Z, Pi S, and Pi SZ phenotypes but varied in their abili...

Measurement of nine human milk proteins by nephelometric immunoassays: application to the determination of mature milk protein profile.

OBJECTIVES Microparticle-enhanced nephelometric immunoassays for six human milk proteins (beta-casein, kappa-casein, alpha-lactalbumin, serum albumin, lactoferrin, and lysozyme) and conventional immunonephelometry assays for immunoglobulin A, C3, and C4 complement proteins were developed and characterized. DESIGN AND METHODS Microparticle-enhanced nephelometric immunoassays are competitive as...

A new High Sensitive Functional Nephelometrical Assay for Assaying C- reactive protein in Serum Based on Phosphocholine Interaction

C-reactive protein (CRP) is able to bind phosphocholine in the presence of calcium ions. According to a previous functional property of CRP, we tried to develop an affordable and cheap high sensitive nephelometric CRP assay using soy oil. Serum samples were measured by Nephelometer BNII (Siemens), by mixing the serum with diluted soy oil emulsion (Intralipid ® 20%) and Tris-calcium buffer (PH 7...

A rate nephelometer for measuring specific proteins by immunoprecipitin reactions.

A kinetic nephelometric method and instrument have been developed for the rapid determination of specific serum proteins by means of immunoprecipitin reactions. The maximum rate of change of scattered light intensity in an antigen-antibody reaction can be made to occur within 60 s after initiation of the reaction and provides a measure of the antigen concentration under antibody excess conditio...

Use of laser nephelometry in the measurement of serum proteins.

Measurement of immunoglobulins (Ig) and of complement component C3 in human serum by automated and manual nephelometric techniques is tedious, and the effective linear range is too narrow. We describe a laser nephelometer/reagent system for measuring serum proteins by the use of forward light scatter (which enhances the ratio of reaction/blank), electronic blank subtraction, laser light (632 nm...
